Abstract :

Kashin-Beck disease (KBD) is a childhood-onset endemic osteoarthropathy in China. Nutrients including trace elements may play active roles in the development of KBD.This study aimed to estimate the nutrient intakes of children in endemic areas and to identify the imbalanced nutrients associated differentially expressed genes in the juvenile patients with KBD.In this cross-sectional study, a consecutive 3 day 24 h semi-quantitative dietary retrospect questionnaire was conducted to estimate the daily nutrient intakes of children using CDGSS 3.0 software. Gene profile analysis was employed to identify differentially expressed genes in peripheral blood mononuclear cells of children with KBD. GOC, CTD, KEGG, and REACTOME databases were used to establish the relationship between nutrients and nutrients-associated differentially expressed genes and pathways. Statistical analyses were accomplished by SPSS 18.0 software.Daily Se intakes without supplementation of children were significantly lower in Se-supplemented (Se + ) KBD areas (29.3 ∼ 29.6 mg/d) and non-endemic area (27.8 ± 7.9 mg/d) compared to non-Se-supplemented (Se-) KBD area (32.9 ± 7.9 mg/d, c<sup>2</sup> = 20.24, P < .01). Children in Se+ KBD areas were suffering more serious insufficient intake of multiple nutrients, including vitamins-B2/-C/-E, Ca, Fe, Zn and I. Gene profile analysis combined with bioinformatics technique identified 34 nutrients associated differentially expressed genes and 10 significant pathways which are related to the pathological changes in juvenile KBD.Imbalance of dietary nutrients and nutrients-associated differentially expressed genes and pathways may play important roles in the development of juvenile KBD.

Selection of Implantation Sites for Transplantation of Encapsulated Pancreatic Islets. PubMed

Abstract :

Pancreatic islet transplantation has been validated as a valuable therapy for type 1 diabetes mellitus patients with exhausted insulin treatment. However, this therapy remains limited by the shortage of donor and the requirement of lifelong immunosuppression. Islet encapsulation, as an available bioartificial pancreas (BAP), represents a promising approach to enable protecting islet grafts without or with minimal immunosuppression and possibly expanding the donor pool. To develop a clinically implantable BAP, some key aspects need to be taken into account: encapsulation material, capsule design, and implant site. Among them, the implant site exerts an important influence on the engraftment, stability, and biocompatibility of implanted BAP. Currently, an optimal site for encapsulated islet transplantation may include sufficient capacity to host large graft volumes, portal drainage, ease of access using safe and reproducible procedure, adequate blood/oxygen supply, minimal immune/inflammatory reaction, pliable for noninvasive imaging and biopsy, and potential of local microenvironment manipulation or bioengineering. Varying degrees of success have been confirmed with the utilization of liver or extrahepatic sites in an experimental or preclinical setting. However, the ideal implant site remains to be further engineered or selected for the widespread application of encapsulated islet transplantation.

A Reduced GO-Graphene Hybrid Gas Sensor for Ultra-Low Concentration Ammonia Detection. EI PubMed Scopus

Abstract :

A hybrid structure gas sensor of reduced graphene oxide (RGO) decorated graphene (RGO-Gr) is designed for ultra-low concentration ammonia detection. The resistance value of the RGO-Gr hybrid is the indicator of the ammonia concentration and controlled by effective charge transport from RGO to graphene after ammonia molecule adsorption. In this hybrid material, RGO is the adsorbing layer to catch ammonia molecules and graphene is the conductive layer to effectively enhance charge/electron transport. Compared to a RGO gas sensor, the signal-to-noise ratio (SNR) of the RGO-Gr is increased from 22 to 1008. Meanwhile, the response of the RGO-Gr gas sensor is better than that of either a pristine graphene or RGO gas sensor. It is found that the RGO reduction time is related to the content of functional groups that directly reflect on the gas sensing properties of the sensor. The RGO-Gr gas sensor with 10 min reduction time has the best gas sensing properties in this type of sensor. The highest sensitivity is 2.88% towards 0.5 ppm, and the ammonia gas detection limit is calculated to be 36 ppb.

Abstract :

The expected corresponding increase in tonsillar human papillomavirus (HPV) infection associated with increasing incidence of oropharyngeal squamous cell carcinoma (OPSCC) substantiate the evaluation of normal tonsillar tissue in different population. The epidemiology of HPV in tonsillar tissue varies geographically. This study evaluated samples from two countries to determine the prevalence in respective samples.To characterize HPV infection in non-malignant tonsillar tissue from Shaanxi, China (herein after referred to as China) and Punjab, Pakistan (herein after referred to as Pakistan).The DNA extracted from formalin-fixed, paraffin-embedded tumor free tonsillar tissue was evaluated using polymerase chain reaction (PCR). A total of 367 cases from China and 139 cases from Pakistan were screened for HPV DNA using GP5+/GP6+ consensus primer. Genotype of the positive cases was determined for common HPV types (6, 11, 16, 18, 52, 58) simultaneously by type-specific PCR.The mean age of cohorts in China was 13.42 (Median age 7, Range 2-72 years) while in Pakistan it was 10.77 (Median age 8, Range 3-42 years) the gender distribution was 61.6% male in China and in Pakistan they were 56.8%, rest were females. The overall prevalence of HPV in China was 2.45% and 2.16% in Pakistan. High risk human papillomavirus was 1.63% in China with 5 cases positive for HPV 16 and HPV 58 in 1 case. In Pakistan, 2 cases (1.43%) of HPV 16 were detected. Low-risk types include HPV 11 present in 2 cases from China, while HPV 6 was detected in 1 case each from both the countries.A low prevalence of HPV was found in China and Pakistan; high-risk and low-risk HPV were detectable in tonsillar tissue from both countries. Age stratification (< 5 years, 5-14 years, 15-25 years, > 25 years) suggest that sexual and non-sexual transmission of the virus can occur. The difference in the genotype distribution geographically within China and with Pakistan was observed in the case of HPV 58. The most common type in both the countries was HPV 16.

Abstract :

Colorectal neoplasia differentially expressed (CRNDE) served as an oncogenic long noncoding RNA (lncRNA) to be involved in the initialization and development of human cancers. However, the clinical significance and biological function of CRNDE in clear cell renal cell carcinoma (ccRCC) was not fully understood. In our study, we found CRNDE levels were increased in ccRCC tissue specimens and cell lines, and corrected with advanced clinical stage, large tumor size, lymph node metastasis, distant metastasis, and poor pathological grade in patients with ccRCC. Furthermore, levels of CRNDE were negatively correlated with overall survival of patients with ccRCC, and high-expression of CRNDE was an independent poor prognostic factor for patients with ccRCC. Moreover, loss-of-function and gain-of-function approaches showed CRNDE-enhanced ccRCC cell migration and invasion through modulating EMT-associated genes. In conclusion, CRNDE acts as an oncogenic lncRNA in ccRCC.

Abstract :

A biliary contrast agents pushing device, including a syringe pushing system and a remote controller is introduced. The syringe pushing system comprises an injector card slot, a support platform and an injection bolus fader. A 20 mL syringe can be fitted on the syringe pushing system and kept with the ground about 30 degree. This system can perform air bubble pumping back and contrast agents bolus injection as well as speed adjustment. Remote controller is an infrared remote control which can start and stop the syringe pushing system. With this device, the remote controlled cholangiography technology can be achieved, which can not only protect doctors from X-ray radiation but also improve the traditional T-tube cholangiography and the contrast effect, reduce postoperative complications in patients as well. The application of this device will improve the current diagnosis and treatment system, the device will benefit the majority of doctors and patients.

Biomarkers are urgently required for predicting rejection so that anti-rejection treatment can be taken early to protect the allograft from irreversible damage. We hypothesized that the combination of circulating fractalkine, IFN-γ and IP-10 might serve as effective biomarkers for predicting early acute renal allograft rejection. We conducted a retrospective study of 87 subjects, who were classified into acute rejection group (ARG; n = 38) and non-rejection group (NRG; n = 49). Serum fractalkine, IFN-γ and IP-10 levels were measured by Luminex. The levels of fractalkine on day 0 and 7th day, IP-10 on 4th and 7th day, and IFN-γ on 7th day in ARG was significantly higher than that in NRG. Kaplan-Meier survival analysis highlighted the higher-levels groups of fractalkine on day 0, 4th and 7th day, IFN-γ on day 0, 1st, 4th, and 7th day and IP-10 on the 4th and 7th day in rejection-free survival probability were significantly lower than low-levels groups. ROC analyses highlight the superiority of fractalkine on day 0, IP-10 on day 0, 4th and 7th day, and IFN-γ on day 0, 1st and 7th day in prediction of acute rejection. We found the combination of fractalkine on day 0, IP-10 on 7th day and IFN-γ on 7th day had the highest AUC (0.866) for predicting rejection with a sensitivity of 86.8% and a specificity of 89.8%. Our findings demonstrated a more powerful prediction of early acute renal allograft rejection during the first month after transplantation by combination of multiple-biomarkers of fractalkine, IFN-γ and IP-10, and the results might help stratify the immunologic risk of acute allograft rejection in recipients.

Functional expression of olfactory receptors using cell-free expression system for biomimetic sensors towards odorant detection. EI PubMed Scopus

Abstract :

How to obtain sufficient functional olfactory receptors and coupled to transducers with high efficiency are crucial to biomimetic olfactory-based biosensors. In this study, a cell-free expression system was employed to prepare functional olfactory receptors that were utilized as sensitive elements for biomimetic olfactory receptor-based biosensors. A nematode olfactory receptor, ODR-10, was used as a model of olfactory receptors and expressed in Escherichia coli (E. coli) cell-free expression system. To improve the coupling efficiency and obtain the on-chip purification, a His6-tag was expressed as a fusion to ODR-10, which makes the expressed ODR-10 capable of selectively binding to the sensor surface modified with anti-His6-tag aptamers. Electrolyte-insulator-semiconductor (EIS) sensors were utilized as the transducer to measure the capacitance changes induced by the ODR-10 responding to its natural ligand, diacetyl. The results show that ODR-10 was successfully expressed using E. coli cell-free expression system and the expression could be promoted by adding 0.5% Brij58 as the detergent. Capacitance measurement results indicate that this olfactory receptor-based biosensor can detect diacetyl with high specificity and sensitivity. Concentration-dependent linear response to diacetyl ranging from 0.01 nM to 1 nM was obtained. The detection limit was as low as 0.01 nM. All the results demonstrated that the cell-free expression system provides a new approach for the preparation of functional olfactory receptors, which have great potential to be applied in biomimetic sensors towards chemical sensing.

Recently, extended mixed dislocations were observed at a [001]/(100) low-angle tilt grain boundary of a SrTiO3 bicrystal because of a slight twist between the two crystal parts. The b = a[201]/(100) mixed dislocations at the grain boundary dissociate into three dislocations with Burgers vector b of a/2[101], a[100], and a/2[101], respectively. A structure model has been proposed in particular for the dislocation cores of the two partials with b = a/2[101] based on the high-angle annular dark-field (HAADF) images acquired by scanning transmission electron microscopy (STEM). However, the details of the atomic structure and the chemical composition of the dislocation cores remain unexplored, especially for the b = a[100] dislocation that is evidently disassociated into two b = a/2[101] partial dislocations. In this work, we study the further atomic details of the extended mixed dislocations, in particular the local chemistry, in a SrTiO3 bicrystal using STEM, electron energy loss spectroscopy (EELS), and energy dispersive X-ray (EDX) spectroscopy techniques. By these atomic-scale imaging techniques, we reveal a unique feature for the atomic structure of the b = a[201]/(100) extended mixed dislocation, which we named as local crystallographic shear (LCS) structures. In addition, we identify a rock salt FCC-type TiOx (x = 0.66-1.24) phase at the locations of the extended mixed dislocations. In contrast to the insulating TiO2 phases, the TiOx phase is known to exhibit very low electrical resistivity of only several μΩ cm. In this regard, the extended mixed dislocations of SrTiO3 comprising the FCC TiOx phase may function as the conducting filament in resistive switching processes by completion and disruption of the TiOx phase along the dislocation cores through electrically stimulated redox reactions.

<b>Objective:</b> To understand the effect of interpregnancy interval (IPI) of childbearing aged women on the birth weight of single live birth neonates in Shaanxi province. <b>Methods:</b> A questionnaire survey was conducted among the childbearing aged women selected through multistage stratified random sampling in Shaanxi during 20l0-2013, all the childbearing aged women had definite pregnancy outcomes. The interpregnancy interval of the childbearing aged women and the birth weight of the newborns were used as the independent variables and dependent variables respectively in multiple linear regression model and quantile regression model, and confounding factors were controlled. <b>Results:</b> A total of 13 063 women at childbearing age and their infants were investigated. The incidence of low birth weight and macrosomia was 3.54% and 7.62% respectively. Multiple linear regression analysis showed that there was no significant difference in birth weight among different IPI group and control group. Quantile regression analysis showed when birth weight was at 5 percentiles, the difference in birth weight between newborns whose mothers had short interpregnancy interval (<12 months) and those in control group was significant, when the birth weight was ≥90 percentiles, the birth weights of newborns whose mothers had long interpregnancy interval (60-119 months) were higher than those in control group, the difference was significant. As the increase of the percentiles of birth weight, the extent of changes gradually increased. <b>Conclusion:</b> The analysis indicated that both short (<12 months) and long (60-119 months) interpregnancy interval would had negative effects on birth weights of newborns. Therefore, planning for pregnancy is important for having a good perinatal outcome.<b>目的:</b> 探讨陕西省育龄妇女生育间隔对活产单胎新生儿出生体重的影响。 <b>方法:</b> 采用分层多阶段随机抽样方法,以调查问卷方式收集2010-2013年陕西省怀孕结局明确的育龄妇女及其子女的相关信息。将生育间隔和新生儿出生体重分别以自变量和因变量纳入多重线性回归及分位数回归模型,并控制相关混杂因素。 <b>结果:</b> 共调查13 063名育龄妇女及其子女,低出生体重和巨大儿的发病率分别为3.54%和7.62%。多重线性回归结果显示,不同生育间隔分组与参照组相比,新生儿出生体重差异无统计学意义。分位数回归结果显示,当出生体重处于第5百分位点时,生育间隔短(<12个月)与参照组相比出生体重差异有统计学意义;当出生体重处于第90百分位点及以上时,生育间隔长(60~119个月)者其新生儿出生体重高于参照组,差异有统计学意义,且随着出生体重的增加,提高的幅度随之增加。 <b>结论:</b> 母亲生育间隔过长或过短均可对新生儿出生体重造成不良影响,因此计划怀孕对获得良好的围生结局具有重要意义。.
