考察了几种经过化学改性后带有离子交换配基的琼脂糖凝胶介质的理化性能.通过外形观察、元素及金属含量分析、柱效测定、耐压及流速测定、离子交换容量和蛋白吸附容量测定,从多方面对性能进行验证比较,并对其在分离纯化重组人血白蛋白的应用性能进行了初步研究.结果表明国产产品与进口产品理化性能相当.

Abstract :

beta(2)-Adrenoceptor (beta(2)-AR) was purified from the rabbit lung tissue by sepharose-salbutamol affinity chromatographic column. To prepare the beta(2)-AR stationary phase, beta(2)-AR was evenly immobilized on the surface of macro-pore silica with a mild chemical coupling method through covalent bond. The retention properties of beta(2)-AR stationary phase were characterized by four ligands, salbutamol sulfate, noradrenaline bitartrate, adrenaline hydrochloride and propranolol hydrochloride, to establish the beta(2)-AR affinity chromatography. Then, the method was used to screen the active compounds from the total extracts of Semen Armeniacae Amarum. The results showed that beta(2)-AR on the surface of the stationary phase could keep its original bioactivity and selectivity. Amygdalin retained in the chromatographic column was proved to be the active compound of the total extracts of Semen Armeniacae Amarum. Compared with the existing chromatographic screening approaches, this method showed a good stability and high selectivity. The active compounds which could interact with beta(2)-AR in traditional Chinese medicine (TCM) could be screened efficiently by this method, providing a new way to screen the active compounds in complicated samples such as TCM.

Vasorelaxant effect of isopropyl 3-(3,4-dihydroxyphenyl)-2-hydroxypropanoate, a novel metabolite from Salvia miltiorrhiza, on isolated rat mesenteric artery SCIE Scopus

The present study was designed to investigate the relaxant effect of isopropyl 3-(3, 4-dihydroxyphenyl)-2-hydroxypropanoate (IDHP), a new metabolite from Salvia miltiorrhiza, on rat mesenteric artery. Isolated mesenteric arterial rings were mounted in organ baths and the isometric tension changes were measured continuously by a sensitive myograph system. The results showed that IDHP at concentrations greater than 0.1 nM produced a concentration-dependent relaxation of artery contracted by norepinephrine with pEC(50) of 7.41 +/- 0.08. Removal of the endothelium did not affect this relaxation, suggesting that IDHP exerted a direct effect on vascular smooth muscle cells. Meanwhile, the vasorelaxant effect of IDHP was unaffected by pre-treatment with ATP-sensitive K+ channel inhibitor glibenclamide, delayed rectifier K+ channel inhibitor 4-aminopyridine, inwardly rectifying K+ channel inhibitor barium chloride and beta-adrenoceptor antagonist propranolol. However, the non-specific K+ channel inhibitor tetraethylammonium (TEA, 3 mM) produced a rightward shift of 1.8 fold on the concentration-response curve of IDHP. Moreover, IDHP shifted the concentration-response curve of CaCl2 as well as two receptor-mediated constrictors, phenylephrine and 5-hydroxytryptamine, to the right in a non-parallel manner. In the absence of extracellular Ca2+, IDHP depressed the contractions induced by norepinephrine and CaCl2, and the maximal inhibitions were 48.3 +/- 18.9% and 58.4 +/- 10.9%, respectively. These results suggest that IDHP exerts a vasorelaxant effect by inhibiting both Ca2+ release from intracellular stores and Ca2+ influx through voltage-dependent calcium channels, and receptor-operated calcium channels in vascular smooth muscle cells. In addition, activation of vascular TEA-sensitive K+ channels may be partially involved in the relaxant effect of IDHP. (c) 2007 Elsevier B.V. All rights reserved.

自制填充介质对人血浆凝血因子Ⅸ的层析分离 CSCD

目的 采用自制的DEAE Bio-Sep FF和肝素Bio-Sep FF介质,从人血浆中快速分离纯化凝血因子Ⅸ(FⅨ).方法 低温离心去除冷沉淀后的人血浆,在不同淋洗条件下,经过两步弱阴离子交换和一步亲和层析分离纯化FⅨ,用活性检测试剂盒检测各组分凝血因子的活性,Bradford法测定蛋白浓度,聚丙烯酰胺凝胶电泳分析样品的纯度.结果 经过三步层析分离,得到的FⅨ比活达到99.40 IU/mg,纯化倍数为3 823倍,回收率约为30%,纯度较高.结论采用该方法 和介质,可从人血浆中纯化FⅨ,且效果较好.

Abstract :

从多个方面测定、比较Bio-sep CL 6B与Sepharose CL-6B两种商品化交联琼脂糖凝胶的性能.采用不同分辨率显微镜观察凝胶外观形态、进行耐热试验、分析有机元素及金属元素含量,以及测定二者耐压、流速、色谱特性等.结果表明,两种产品外观均为乳白色球状颗粒,球形形态完好,粒径范围在30~150 μm具有较好的分布;均可耐受120 ℃下30 min热处理;有机元素碳、氢、氮以及其它微量金属元素含量相近;两种产品最大耐压都在80 kPa以上,最大流速可达7.0 mL/min.分离特性测定表明,两种产品色谱分离特性基本相同,对分子量在104~106道尔顿之间的蛋白能实现较好的分离.

A fast solid phase extraction-high performance liquid chromatography-ion trap mass spectrometry (SPE-HPLC-MSn) method is presented for quantitative determination of 3-(3,4-dihydroxyphenyl)-2-hydroxylpropionic acid (Danshensu, DSS) from Compound Danshen Dripping Pills (CDDP) in rabbits' heart, liver, kidney, and lung and for qualitative analysis of the bioactive metabolites of Radix Salvia miltiorrhizae (DS) from CDDP in these tissues. The proposed method was validated with a detection limit of 0.06 ng/mL and a linear range of 5.0 - 200 ng/mL for DSS. The main metabolites of DS were pyrocatecbol, vanillic acid, isopropyl 3-(3,4-dihydroxyphenyl)-2-hydroxypropanoate, and 2-hydroxyl-3-(3,4-dihydroxyphenyl)propinonic sulfuric anhydride. M vitro pharmacological experiments demonstrated that isopropyl 3-(3,4-dihydroxyphenyl)-2-hydroxypropanoate had a distinct vasodilatating action.

Aim: To study the influence of Santalum album (Shi herb) on the pharmacokinetics of Radix Salvia miltiorrhizae (Jun herb) in the rabbits receiving Danshenyin. Methods: A valid assay for the determination of Danshensu was established by HPLC-ESI-MS/MS. The plasma was pretreated with 10% perchloric acid and the supernatant was extracted by ethyl acetate. The p-hydroxybenzoic acid was used as an internal standard to detect the blood-drug concentrations by HPLC-ESI-MS/MS. The collected data were processed by computer program DAS to study the effects of Santalum album on the pharmacokinetics of Radix Salvia miltiorrhizae. Results: The linear range of Danshensu in the assay was 0.2-40.0 μg/ mL. The limit of detection (LOD) was 1.0 ng. The RSD as reproducibility in experiment was 1.34% (n = 6). The average recovery at lower, middle, and upper spiked concentrations was (74.21 ± 3.58)%, (95.5 ± 2.81)% and (80.69 ± 3.93%), respectively. Moreover, prior to and post-oral dosing of compatibility, the pharmacokinetics of Danshensu was found to be fitted to open two-compartment model. And co-administration resulted in obvious increase in AUC0-∞. Conclusion: Shi herb could promote the absorption of Jun herb in Danshenyin. The results could provide clues for understanding of pharmacological mechanism of Jun-Shi pair herbs.

目的建立全面控制葛根及其制剂的定性和定量分析方法.方法用反相高效液相色谱法(RP-HPLC),以甲醇-水(42∶58)为流动相在250 nm波长下检测,对葛根有效部位(醋酸乙酯提取部位)进行指纹图谱定性分析,对葛根有效部位中的有效成分葛根素和大豆苷元进行定量测定,并对11种不同来源的葛根药材进行分析测定.结果在定性分析条件下,葛根有效部位的HPLC指纹图谱中有5个共有峰,可以作为其定性鉴别的指标峰;在以葛根素和大豆苷元为定量指标分析中,其方法的回收率分别为98.0%和95.4%,相对标准偏差(RSD)分别为1.30%和1.61%,葛根药材中葛根素和大豆苷元的含量分别在0.213%~3.626%和0.012%~0.049%范围内.结论以葛根有效部位指纹图谱定性分析和有效成分定量分析方法,可以准确有效地控制其质量.

Compound Danshen Dripping Pill (CDDP) is an important drug widely used for the treatment of cardiovascular diseases. An active component Danshensu (DS) of CDDP was separated by reversed-phase high performance liquid chromatography using column-switching system and analyzed by electrospray mass spectrometry. With this validated assay the pharmacokinetics of CDDP was studied in 10 healthy volunteers after a single oral administration of 250 mg. After trichloroacetic acid precipitation of serum proteins, the analytes were preconcentrated and black-flushed on a reversed-phase column for separation using a switching valve. The analytes were ionized using negative electrospray ionization (ESI) mode. The precursor ion of m/z 196.6 was used to quantify DS in serum. The linear calibration curve ranged from 1.25 to 175mug/mL. The limit of quantification (LOQ) for DS was 0.15mug/mL. The intra-day and inter-day precision (R.S.D.) was less than 7.4 and 7.9%, respectively. (C) 2004 Elsevier B.V. All rights reserved.
