Abstract:
Axially chiral compounds are valuable chiral compounds used as core skeleton of natural products, pharmaceutical intermediates and chiral ligands. Compared to asymmetric couplings catalyzed by metal catalysts or other asymmetric chemical methods, biocatalytic methods are mild, environmental friendly and highly selective. With the fast development of enzyme engineering and directed evolution, biocatalytic asymmetric synthesis of axially chiral compounds has attracted increasing research interest. This article reviews major progress in the area of biocatalytic synthesis of axially chiral compounds starting from different synthetic methods kinetic resolution (KR), dynamic kinetic resolution (DKR) and desymmetrization, and discusses the prospects of applications and existing problems of this area. © 2021, Editorial Board of 'Journal of Chemical Engineering of Chinese Universities'.
