Abstract :

Pyroptosis is a novel manner of cell death that can be mediated by chemotherapy drugs. The awareness of pyroptosis is significantly increasing in the fields of anti-tumor research and chemotherapy drugs. Invoking the occurrence of pyroptosis is an attractive prospect for the treatment of lung cancer. Here, the compound L61H10 was obtained as a thiopyran derivative to compare its activity with curcumin. It was indicated that L61H10 exhibited good anti-tumor activity both in vitro and in vivo via the switch of apoptosis-to-pyroptosis, which was associated with the NF-κB signaling pathway. In addition, L61H10 had no obvious side effects both in vitro and in vivo. In brief, L61H10 is shown to be a potential anti-lung cancer agent and research on its anti-tumor mechanism provides new information for chemotherapy drug research.

Abstract :

A novel method was developed for determination of aflatoxin B1, B2, G1, and G2 (AFB1, AFB2, AFG1, and AFG2) in wheat using inkjet-based dispersive liquid-liquid microextraction (DLLME) coupled with ultrahigh-pressure liquid chromatography-tandem mass spectrometry. A drop-on-demand jetting device was used to form a cloudy solution in traditional DLLME by injecting extraction solvent (10 μL) as ultrafine droplets (∼20 μm diameter) at high frequency into sample solution. The method was validated using wheat as a representative matrix, which was pretreated with acetonitrile/water solution. Good linearity was observed over the studied range (0.06-6 μg/kg), and the limits of quantification (0.06-0.18 μg/kg) were below the maximum level established by the European Union for cereal. Satisfactory recoveries, ranging from 83.2% to 93.0% with relative standard deviations below 4.6%, were obtained for all compounds. The method, which is convenient and reliable and has low solvent consumption, represents a new direction for the development of traditional DLLME technology.

Abstract :

The biomimetic functionalization of carbon nanotubes (CNTs) are promising for its application in solid phase extraction because they can overcome current limitations of covalent functionalization and obtain desired specific recognition ability. The present study aimed to develop a novel dual functionalization CNTs modified with magnetic nanoparticles and high alpha(1A)-adrenergic receptor (alpha(1A)-AR) expression HEK 293 cell membrane. On the basis of the specific interactions between drugs and receptors on cell membrane, high alpha(1A)-AR expression cell membrane camouflaged magnetic CNTs (CMMCNTs) were employed as a drug discovery platform to screen potential alpha(1A)-AR antagonists from traditional Chinese medicine (TCM). The resultant CMMCNTs had a right-side-out membrane and desired magnetic property, enabling to rapidly target and separate the bioactive compounds from complex samples. Furthermore, the CMMCNTs possessed high adsorption capacity, fast binding kinetics, and satisfactory selectivity. As a result, two bioactive compounds as lappaconitine and benzoylmesaconine were screened and identified in the actual application. Moreover, the lappaconitine was validated to have favorable antagonism to alpha(1A)-AR through preliminary pharmacological assays. Hence, this successful application of CMMCNTs showed significant advantages in drug discovery so as to drive the development of TCM. More importantly, the cell membrane biomimetic modification could be widely used to improve the performance of CNTs in various applications.

Abstract :

Rapid and accurate identification of multiple toxins for clinical diagnosis and treatment of mushroom poisoning cases is still a challenge, especially with the lack of authentic references. In this study, we developed an effective method for simultaneous identification of amanita peptide toxins by liquid chromatography coupled with photodiode array detection and ion trap time-of-flight mass spectrometry. The accuracy and selectivity of the methodology were validated through similar multiple fragmentation patterns and characteristic ions of standard alpha- and ss-amanitin. The developed method could successfully separate and identify major toxic constituents in Amanita mushrooms. Two amatoxins and three phallotoxins were confirmed in a single run through their fragmentation patterns and characteristic ions, which can be used as diagnostic fragment ions to identify mushroom toxins in complex samples. Furthermore, the performance of the developed method was verified by using real biological samples, including plasma and urine samples collected from rats after intraperitoneal administration of toxins. Thus, the development methodology could be crucial for the accurate detection of mushroom toxins without standard references.

Abstract :

Exogenous supplementation of antioxidants with ROS scavenging activity would be a potential therapy to cerebral ischemia-reperfusion injury in stroke. In the present study, a series of NDGA analogues with attenuation oxidative stress by directly scavenging ROS and indirectly through keap1/Nrf2/ARE pathway activation were designed and synthesized. All analogues were found to effectively remove ROS directly by DPPH radical scavenging assay, and compound 3a conferred potent protection from the oxidative injury in PC12 cells via promoting Nrf2 to translocate into nucleus and increasing expression of heme oxygenase-1(HO-1), where strongly reduced intracellular ROS level indirectly. More importantly, 3a significantly reduced brain infarction after cerebral ischemia-reperfusion injury in rats subjected to transient middle cerebral artery occlusion (MCAO). Overall, our findings shown compound 3a could serve as a promising compound for the treatment of stroke. (C) 2017 Published by Elsevier Masson SAS.

Abstract :

A rapid and precise liquid chromatography coupled with hybrid ion trap/time-of-flight mass spectrometry method to detect and quantify caulophine and its possible active metabolites in rat plasma and urine was developed. Samples were prepared by plasma protein precipitation combined with a liquid-liquid extraction method. The separation was carried out on an InertSustain (R) C18 column with a mobile phase comprising methanol and 0.1% aqueous formic acid solution. The analysis was complete in 20 min with a flow rate of 0.4 mL/min. Taspine was used as the internal standard. Mass spectrometric detection was conducted with hybrid ion trap/time-of-flight equipped with electrospray ionization in the positive ion mode. The calibration curves of caulophine were linear over the concentration ranges of 0.002-0.20 mu g/mL for plasma and 0.005-0.50 mu g/mL for urine with the correlation coefficients greater than 0.998 in both cases. The method was successfully used to investigate the pharmacokinetics and bioavailability in rat plasma and urine samples after intragastric and intraperitoneal administration of caulophine sodium salt.

Coptidis Rhizoma binds to the membrane receptors on hPDLSC/CMC, and the active ingredient Berberine (BER) that can be extracted from it may promote the proliferation and osteogenesis of periodontal ligament stem cells (hPDLSC). The membrane receptor that binds with BER on the cell surface of hPDLSC, the mechanism of direct interaction between BER and hPDLSC, and the related signal pathway are not yet clear. In this research, EGFR was screened as the affinity membrane receptor between BER and hPDLSC, through retention on CMC, competition with BER and by using a molecular docking simulation score. At the same time, the MAPK PCR Array was selected to screen the target genes that changed when hPDLSC was simulated by BER. In conclusion, BER may bind to EGFR on the cell membrane of hPDLSC so the intracellular ERK signalling pathways activate, and nuclear-related genes of FOS change, resulting in the effect of osteogenesis on PDLSC.

Aiming at improving reliability and tedious analysis time in conventional cell membrane chromatography, an improved bioaffinity sample pretreatment technique with enhanced stability was developed to fast screen and extract potential allergenic components from traditional Chinese medicine injections. In this study, rat basophilic leukemia-2H3 cell membrane coated silica particles (RBL-2H3/CMCSPs) were fabricated by irreversible adsorption between the cell membrane and silica and self-fusion of the cell membrane, which could simulate drug-receptor interactions in vitro. Also, benefiting from the use of paraformaldehyde, the average recoveries of the six batches of RBL/CMCSPs were 90.2% with a relative standard deviation of less than 7.8%, showing that the stability of the materials was remarkably improved compared to materials without fixation. After the successful characterization by spectroscopic and imaging instruments, the prepared RBL-2H3/CMCSPs exhibited desirable adsorption capacity and selectivity. The RBL-2H3/CMCSPs combined with high performance liquid chromatography coupled with time of flight mass spectrometry were then successfully applied to screen and identify two potential allergenic components from huangqi injection, the allergenic activities of which were further investigated by beta-hexosaminidase release assay and histamine release assay in vitro. Overall, this work provides a good platform for the fabrication of bioaffinity sample pretreatment materials with high stability and a long lifespan, which can be a time-saving and energy-saving bioaffinity method to rapidly screen and preconcentrate target compounds from complex samples.

IntroductionCell membrane chromatography (CMC), as a highly selective type of affinity chromatography, has been demonstrated as an effective method to screen bioactive components acting on specific receptor from a complicated biological system. ObjectiveTo review the recent research progress and the technical applications of these analytical methods using CMC combined with gas chromatography-mass spectrometry, (GC/MS) and liquid chromatography-mass spectrometry (LC/MS). MethodologyIn this review, we briefly introduce the CMC offline GC/MS, CMC online GC/MS, CMC offline LC/MS, and CMC online LC/MS system. And the practical application of these technologies is also enumerated. Then the future of these technologies and research methods were discussed. ResultsMany bioactive components interacting with specific receptors have been screened and identified in traditional Chinese medicines. ConclusionCMC technique has been combined with GC/MS and HPLC/MS and these combined systems have been successfully used to screen bioactive components acting on specific receptors from a complicated biological system. We briefly introduce the CMC offline GC/MS, CMC online GC/MS, CMC offline LC/MS, and CMC online LC/MS system. And the practical application of these technologies is also enumerated. Diosgenin wastewater produced from Dioscorea zingiberensis C. H. Wright in diosgenin enterprise is the by-product of numerous operations chiefly including washing raw materials, acid hydrolysis, and cleaning the apparatus with water. It requires the removal of high concentrations of various organic and inorganic contaminants from this wastewater in order to avoid pollution to the environment and fresh water system. Currently, several different methods such as chemical, physicochemical, biological recovery resource, constructed wetland, and integrated methods of these processes are being applied for treating diosgenin wastewater. Aiming to get a comprehensive overview, available reports from experimental scale to industry scale about the treatment of this wastewater were collected and summarized. Therefore, this review paper will provide appropriate choice for the efficient treatment of diosgenin wastewater and supportive information for the other scientists who want to continue this valuable research in the future.
